Consider their dress sense
Create their idiolect
Read their dialogue aloud - is it consistent?
Put them in different situations and see how they respond
Think about how your readers will view them - will they love them, hate them, route for them? Any and all of these are fine because it means they care and will want to know how your protagonist's life turns out at the end of the book.
